IMFINZI mechanism of action for CASPIAN regimen

IMFINZI (durvalumab), a human monoclonal antibody targeting PD-L1, is a key component of the CASPIAN regimen, which incorporates etoposide and either carboplatin or cisplatin for patients with ES-SCLC. By binding to PD-L1, IMFINZI inhibits its interaction with PD-1 and CD80, helping to restore T-cell function and facilitate immune-mediated recognition of tumour cells. This mechanism addresses the immunosuppressive environment commonly observed in ES-SCLC, which can limit the ability of the immune system to mount an effective antitumour response.3

How IMFINZI works for ES-SCLC: a detailed mechanism of action

  • IMFINZI is a human monoclonal antibody that targets PD-L1, a protein overexpressed on tumour and immune cells within the tumour microenvironment3
  • PD-L1 binds to PD-1 and CD80 to reduce T cell activation and suppress immune responses3
  • Tumour cells exploit PD-L1 overexpression to avoid destruction by the immune system3
  • IMFINZI blocks PD-L1 from interacting with PD-1 and CD80, disrupting this immune-evasion mechanism3
  • This blockade enhances cytotoxic T cell activation, restoring their ability to recognise and destroy cancer cells3
